The proposal aims to investigate the role of reduced autophagy in aged cardiomyocytes, possibilities to interfere with a decrease of autophagy and the role of accumulated and damaged material in functional loss of cardiomyocytes in aging. To analyze the inducibility of autophagy in cardiomyocytes, young and aged cells will be stimulated with Ang II, followed by expression analysis of autophagy markers and there functional characterization. Finally, the observed cellular mechanism will be investigated on an organismal level, using a mouse model of premature senescence.
